This laboratory device measures the fluorescence intensity of the fluorescent reagent, i.e. its intensity and wavelength distribution of the emission spectrum in biological or mineral samples, we can find two types: handheld device and a table unit, its sensitivity can be adjusted to specific light wavelengths using filters and depending on what is being studied, so its maintenance is an indispensable process for proper function.
It has a range of interesting applications including chemistry, biochemistry, medicine, environmental monitoring, being used in any biological laboratory, especially for research, in laboratories for the analysis of physiological samples, based on the principle that each chemical compound absorbs or emits light energy of different wavelength.
Recommendations for use of the fluorometer
- Verify that you have the secondary fluorescence filter in place to read.
- In the case of absorbance measurement, verify that it has the appropriate filter, i.e. the interference filter.
- Be careful to press the DEL button, as, if you delete the data, it will not be possible to retrieve it.
- Program the fluorometer for the calculation of the results of qualitative techniques.
- Place the equipment on a flat table free from mechanical vibration.
- It should be in a place where ventilation is adequate but not likely to be subject to excessive dust.
- Avoid placing equipment where it is close to sources of heat such as heating or sunlight.
- Do not put heavy objects on the computer.
- If you are not connected to power for more than 72 hours, your existing data will be lost
- The instrument should be warm before doing any procedures.
- Perform weekly periodic check of wavelength adjustment, when it is suspected to have changed.
- Check 0 and 100% T every time you are going to read and when the wavelength changes.
- The desired wavelength is selected.
Maintenance and conservation of Fluorometers
- Cleaning of the interference and secondary fluorescence filters.
- Cleaning of internal fluorescence patterns.
- Replace the lamps.
- Changing the fuses.
- Cleaning and/or change of infrared filter.
- Any other maintenance or repair operation may only be performed by a Technical Support Specialist.
- Cleaning of the interference and secondary fluorescence filters.
- Clean the filters with an ethanol-sprayed cotton swab when not in use, stored in the filter holder and avoid touching the filter surface.
- Adjust the apparatus to zero using distilled water.
- A known concentration standard is read and the device is adjusted to that concentration.
General cleaning of the Fluorometer
- Turn off the instrument and disconnect the power cord.
- Wipe the inner part of the kit with a swab moistened with ethanol and dry it with another clean cotton swab. Do not leave any cotton on the spot.
- To prevent dust and dirt from entering the equipment, make sure the cart is always inserted and the front door is closed.
- Halogen lamp operation check, if a replacement is required, you must follow the manufacturer’s instructions.
- Cleaning the surface of the instrument.
- Lamp cleaning and condenser.
